Regardless the type of sleep outcomes, gabapentin showed stable efficacy in the treatment for sleep disturbance in patients with medical illness with a relatively high risk of treatment discontinuation and drug withdrawal when used at an average dose of approximately 1,800 mg/day. HORIZANT® (gabapentin enacarbil) is a prescription medicine used to: HORIZANT is not for people who need to sleep during the daytime and stay awake at night. In this study, the authors show that gabapentin enacarbil reduces RLS-associated sleep disturbance. Strengths of the study include the randomized, multicenter, placebo-controlled design, and the use of objective, quantifiable outcome measures. HORIZANT® (gabapentin enacarbil) Extended-Release Tablets are indicated for the treatment of moderate-to-severe primary Restless Legs Syndrome (RLS) in adults. HORIZANT is not recommended for patients who are required to sleep during the daytime and remain awake at night. Study Impact: Participants with occasional disturbed sleep treated with gabapentin had significantly longer sleep duration and greater depth (versus placebo) in response to a phase advance manipulation known to disrupt sleep maintenance. Gabapentin, an α 2 δ subunit ligand of L-type voltage-gated calcium channels, is indicated for the Sleepiness, Dizziness, and Driving Impairment. Horizant may cause dizziness and sleepiness and can affect your alertness or coordination, including the morning after you take your dose. Most studies show that gabapentin improves slow wave sleep (“deep sleep”) and total sleep time. Two small studies showed that gabapentin may help people with primary insomnia and occasional sleep disturbance improve total sleep time and wakefulness in the morning. GEn 600 mg and 1200 mg significantly improved sleep, mood, pain, and quality of life (QoL) versus placebo. The most frequent treatment-emergent adverse events were somnolence and dizziness. Assess efficacy and tolerability of gabapentin enacarbil (GEn) in adults with severe primary restless legs syndrome (RLS). (1) Participants: all included patients were 18 years or older and had/did not have a record of baseline sleep status; (2) intervention: the patients in the treatment group received gabapentin, gabapentin enacarbil, or XP13512 (Gabapentin), and the patients in the control group received placebos with a treatment duration of at least 7 days; (3 Polysomnogram demonstrated that gabapentin enacarbil improved sleep architecture, including shortening stage 1 sleep and extending time spent in slow-wave sleep [Kushida et al. 2009b]. Gabapentin enacarbil (Gen) administered once daily to patients with severe primary RLS significantly improved sleep, mood, pain, and quality of life. Non-dopaminergic drugs such as oral gabapentin (GBP) have been more recently advocated. Despite ameliorating RLS symptoms, GBP’s pharmacokinetic limitations restrict its overall effectiveness. A novel specifically designed prodrug, gabapentin enacarbil (GE), has demonstrated successful RLS alleviation with a superior pharmacokinetic profile. In adults with RLS, the AASM recommends the use of gabapentin enacarbil over no gabapentin enacarbil (strong recommendation, moderate certainty of evidence) 2. In double-blind, multicentre trials, treatment with gabapentin enacarbil 600 mg/day for 12 weeks significantly improved the symptoms of moderate to severe primary RLS in adults. Gabapentin enacarbil also significantly improved RLS pain scores and generally improved sleep and mood outcomes.
